EasySeq™ 18S and Cyp51A rRNA Fungal ID Kit. NGS Library Prep by Reverse Complement PCR


EasySeq™ 18S rRNA and Cyp51A Kit provides a straightforward NGS Library Prep, based on Reverse Complement PCR (RC-PCR). This patented technology combines target amplification and indexing in single PCR tubes, providing a fast, simple and safe workflow for the identification and classification of fungi, including Aspergillus species and Cyp51A mutation (azole resistance) status. Azole resistance among Aspergillus fumigatus strains has increased notoriously. The main resistance mechanisms are relative to point mutations of the azole target, 14-α sterol demethylase (Cyp51A), with or without tandem repeat integrations in the Cyp51A promoter. EasySeq™ is compatible with illumina® NGS sequencing platforms.

With the NimaGen EasySeq™ RC-PCR Library Preparation method, the implementation of an NGS-driven identification and monitoring strategy could not be simpler or safer: minimal hands-on time and the use of a single PCR tube which reduces the risk of error and contamination.

Tiled RC-PCR primers and probes are provided in tubes, as well as breakable 96-well plates which include UDI Indexes. Just add (c)DNA and the RC-PCR mastermix/probe mix to the pre-dispensed Index Plates provided in the kit and load on onto your PCR instrument. Post-PCR all samples can be combined for pooled cleanup with the AmpliClean™ Magnetic Bead Cleanup Kit and following standard quantification and QC checks your libraries are ready to sequence. This workflow solution is ideal for low- to high-throughput automation.
